Identifying circles that have black dots in them

Is there any way to identify all the circles in this image and within that can we identify the circles that have black dots in them?

For example can we identify the circles in red markup that have black dots and circles in blue markup those that dont.

The name of color is for human perception only. Are they absolutely black, if yes, after converting to a gray image, 0 should be a gray level? Please check, or you can check the corresponding coordinates, or 0 in all three color planes.
grayImage=rgb2gray(rgbImage);
data=grayImage==0;
imshow(data);
Next count the all disjoint white blobs.(Simple: See Morpho Operations)
If the level of black gray cannot be considered absolute black, then you can try with a slightly higher gray value.
grayImage=rgb2gray(rgbImage);
data=grayImage<=5; % Check the threshold value
imshow(data);
It is all about putting in more and more effort and achieving accurate results.

Can you again clarify the logic "which cells to detect"?
I am expecting a clear statement for the condition (If Possible: Maths language is much better)
