How can i remove the black color from inside the white curve boundary

what mean this line a=Image1(:,:,1);

Use imfill with the 'holes' option.
% read the image
im = imread('http://www.mathworks.com/matlabcentral/answers/uploaded_files/8143/ar1.jpg');
% binarize it
bw = rgb2gray(im)>100;
% fill holes
out = imfill(bw,'holes');
% display image
imshow(out);

That finds the area of the completely filled white area. Exactly what do you mean when you say "the area of inside the white area". Do you mean the area of the black region that is inside the white area? If so, which one? There are several black regions inside the white region.
