ANOVAで複数の指標を、平均がゼロで分散が未知の正規分布との帰無仮説検定することはできますか
ANOVA の前提は"等分散"なので、分散未知でも問題はありません。また、複数の"指標" が何を指すのかが定かではないですが複数の群に渡って平均が同じであるという帰無仮説を検定します。

式と計画行列の関係が参考になるかと思います。階層構造が相関を表しているとして、A > B > C で、A-B の交互作用、B-C の交互作用をモデル化するのであれば、(* | A:B) のように指定することが出来ます。
