Drawing a line in the matrix of an image directly without using imshow()

I have nearly 300 binary images. I need to draw 12 red lines (whose start and end points are known) on every image directly into the matrix of the image without using imshow(). Please let me know if there is any way doing this?

See this demo to burn a line into the image. First convert your binary image into uint8:
grayImageR = 255 * uint8(binaryImage);
grayImageG = 255 * uint8(binaryImage);
grayImageB = 255 * uint8(binaryImage);
Then it will be straightforward to get just the line part (remove the ellipse part of the demo) and to burn 255 into grayImageR and 0 into grayImageG and grayImageB, then use cat() to turn them into a color image:
rgbImage = cat(3, grayImageR, grayImageG, grayImageB);
