how to identify background taking average from a collection of images

i am having a problem to separate object from a collection of images. those pictures look quite similar still threshold values differ. so i wanted to identify the background taking average from the collection. for the simplicity i am attaching few photos. thanks

Can't you just take a blank shot - one with the slide in there but no cells at all?
