How do I analyze specific points in a binary image?

I am analyzing a cell and trying to determine the differences in position of the cell membrane for multiple frames. I need to measure the entire circular cell at specific points and have those points carry over from frame to frame (over 1000 frames). I am converting the jpeg images to binary to see where it is black and white but I am not sure how to measure the difference in one specific point of the cell.
