How do I analyze specific points in a binary image?

I am analyzing a cell and trying to determine the differences in position of the cell membrane for multiple frames. I need to measure the entire circular cell at specific points and have those points carry over from frame to frame (over 1000 frames). I am converting the jpeg images to binary to see where it is black and white but I am not sure how to measure the difference in one specific point of the cell. Any suggestions or help would be greatly appreciated. Thank you very much. Please feel free to contact me if more information is needed.

It's hard to give image analysis advice without an image to look at. The Crystal Ball Toolbox has not yet been released.
