Represent points on a sphere as quaternion

I need to optimize the attitude maneuver from a given location on earth (lat,lon) to another location given the system dynamics. I am using quaternions to represent the orientation of the satellite but unsure how to set the inital and final orientation of the satellite in terms of the location on the ground. Any suggestions?
